Markram, 23, will replace regular captain Faf du Plessis who on Friday was ruled out of not only the remainder of the ODI series, but the three T20 internationals to follow.
India are currently 1-0 up in the series, with the second match set for Sunday in Centurion.
The first ball is due to be bowled at 10:00.
Congratulations to @AidzMarkram who has been appointed as stand-in captain for the remainder of the @Momentum_za series against India. All the best ???? #SAvIND #ProteaFire pic.twitter.com/BtoMvaEgmd
