Simon, now 33, has a highest ever world ranking of No 6 back in 2009 and has 13 ATP Tour titles to his name.
Harris on Friday beat American JC Aragone 6-4, 6-2 in his third qualifying match to advance to the main draw at Flushing Meadows.
Harris, 21, who hails from Cape Town, earlier in the week defeated Sebastian Ofner from Austria 6-2, 6-1 and Roberto Quiroz of Ecuador 7-6 (7/5), 6-3.
Harris, who is ranked 145th in the ATP Tour world rankings, joins compatriot Kevin Anderson in the main draw.
Anderson, who was runner-up at last year's US Open, will open his campaign against unseeded American Ryan Harrison.
The 2018 US Open will be held from August 27 to September 9.
