Johannesburg - SuperSport United were drawn on Thursday evening against their crosstown rivals University of Pretoria for the Nedbank Cup last 32.
The dates and venues of the matches are yet to be announced.
Matsantsantsa coach Cavin Johnson welcomed the draw and said the Tshwane derby would add another dimension to the clash.
"Just being a derby makes it more exciting but playing Tuks is always a good thing because it brings the best out of your players," Johnson said.
"We are happy with the draw and we are happy with our team going forward."
Johnson said while he did not know when they would face AmaTuks, he was looking forward to the restart of the season.
"We can say we are starting the league early against Pirates on Saturday," he said.
"With that I think we can get some momentum going into the league, cup and Caf much quicker than other teams."
Orlando Pirates got a tricky draw against Thanda Royal Zulu who are fighting relegation in the National First Division and could be looking to use the cup match to find their footing.
"This draw is a bit tough for us because we have such a congested fixture list as you know," said Pirates coach Roger de Sa.
"Obviously another trip all the way to Richards Bay, ideally we would have loved to have had a home draw but that’s how it goes in a cup competition.
De Sa said the team was nevertheless looking forward to the challenge against a desperate Thanda Royal Zulu side.
"I think they are a team that has been struggling this season a little bit from what we have seen but always a tough team," he said.
"Last year we made an early exit against similar opposition in this competition and we can’t make a similar mistake this time around."
Pirates were knocked out in the first round of the Nedbank Cup last season by rookies, Maluti FET College.
Other matches will see Kaizer Chiefs playing away to Black Leopards who are now coached by former Amakhosi coach Kostadip Papic.
