Vakatawa, 27, had heavy strapping on his left arm for a triceps injury during the 24-17 victory over England that gave a new-look France side an encouraging start to their Six Nations campaign last weekend.
The absence of a player who has scored seven tries for club side Racing 92 this season is a blow to France's attacking options.
Stade Francais player Hamdaoui, 25, will miss the game against Italy due to an ankle injury.
