According to BET.co.za, the Springboks are 11/20 favourites to win at the Principality Stadium while Wales, who beat the Boks at the same venue last year, are 7/4 outsiders.
The draw is on offer at 73/4.
In terms of handicap, the Boks are 4.5-point favourites.
The Boks are looking to make it three wins from four on the northern hemisphere tour after they lost to Ireland, but beat rebounded to beat France and Italy.
Wales, meanwhile, have lost to Australia and New Zealand while they scrapped past Georgia.
