The 1999 World Cup Player of the Tournament will replace former New Zealand spinner Daniel Vettori in the position, with the next edition of the BPL taking place between January and February in 2019.
Vettori will instead be working with the Brisbane Heat in Australia's Big Bash League, which runs from December 2018 through top February 2019.
Klusener had previously been the batting coach of the Zimbabwe national team, but he lost that job as part of the clean-out that followed their failure to qualify for the 2019 World Cup.
Klusener began his coaching career in 2012 when he took the head coach role on at the Dolphins, the province he had represented throughout his career.
