The Proteas are 1-0 down in the three-Test series having lost the first Test in Visakhapatnam last week.
If he scores just six runs in the second Test, Du Plessis will become only the third South African to score 2 000 runs as a Test captain.
Graeme Smith comfortably leads that statistic with 8 647 runs while Test skipper with Hansie Cronje second on the list with 2 833.
Du Plessis, who scored 55 and 13 in the first Test match, will captain South Africa for the 31st time in Test cricket on Thursday.
Play on Thursday starts at 06:00 (SA time).
