AmaTuks clinched National First Division honours on Sunday despite losing 3-2 to Mpumlalanga Black Aces.
"This is a great feat by University of Pretoria after what has been an exciting National First Division season," Matthews told the PSL's website.
"I would like to take this opportunity to welcome them to the premier division. There is no doubt in my mind that they have earned this achievement following many years of trying to win promotion.
"Their spirit of perseverance has been rewarded. A lot is expected from them by the football fraternity. Credit should go to Tuks management for ensuring continuity and of course their coach, Steve Barker who has been with this side for a while now. I would also like to welcome Tuks supporters."
Matthews added: "A lot of work lies ahead for them as they prepare for life in the premier division. As the league, we will offer them the best possible support to help them acquaint and adapt better to life in the elite ranks."
