Lambie injured his shoulder early in the Tri-Nations Test against the All Blacks on Saturday.
Frans Steyn is also in doubt after he sustained a hamstring injury against Australia in Durban the previous week.
The Springboks will announce their squad for the Rugby World Cup in New Zealand on Tuesday.
The tournament starts on September 9.
De Villiers was reluctant to answer any questions about the severity of Lambie's injury.
"We have a policy where we discuss injuries on Monday's through our team doctor (Dr Craig Roberts). The group (Bok World Cup group) will be announced on Tuesday and if Pat's name is not there, then you will know that the injury is serious," De Villiers told the Beeld newspaper.
However, the Boks seem a bit more assured over the availability of Steyn.
Steyn is still with the team, which indicates that his hamstring injury is not serious enough for him to miss the World Cup tournament.
