According to the Beeld newspaper, Muller pulled out of the training session with an injured hamstring and the Bok coaches are holding thumbs over his availability for the game against Fiji at the Wellington Regional Stadium on Saturday.
Muller was expected to take the place of the Victor Matfield - who is also out with a sore hamstring suffered against Wales - in the No 5 lock position.
The Springboks' media representative, Andy Colquhoun, could not confirm whether Muller injured the same hamstring that kept him sidelined earlier this year during the South Africa's away leg of the Tri-Nations.
"Towards the end of the training session, Johann felt the some stiffness in his hamstring and as a precautionary measure decided to stop. We will see how he feels on Wednesday and then decide what we need to do," Colquhoun told Beeld.
There was, however, a glimmer of hope for the Boks as their other injured lock, Bakkies Botha, seemed to train without any visible discomfort.
Botha has an injured Achilles tendon and a decision on his availability will also be made on Wednesday.
